NuclearfactorkappaB(NF-kB)transcriptionfactorsareevolutionarilyconserved,coordinatingregulatorsofim-muneandinflammatoryresponses.Theyalsoplayapivotalroleinoncogenesisandmetabolicdisorders.SeveralstudiesduringthepasttwodecadeshavehighlightedthekeyroleoftheIKK/NF-kBpathwayintheinductionandmaintenanceofthestateofinflamma-tionthatunderliesmetabolicdiseasessuchasobesityandtype2diabetes.Recentreports,however,revealanevenmoreintimateconnectionbetweenNF-kBandmetabolism.ThesestudiesdemonstratethatNF-kBreg-ulatesenergyhomeostasisviadirectengagementofthecellularnetworksgoverningglycolysisandrespiration,withprofoundimplicationsbeyondmetabolicdiseases,includingcancer,ageingandanticancertherapy.Inthisreview,wediscusstheseemergingbioenergeticfunc-tionsofNF-kBandtheirsignificancetooncogenesis.
Integrationandcoevolutionofimmuneandmetabolicsystems
NF-kBisbestknownforitsfunctionsinimmunity,inflam-mation,andoncogenesis[1–4].Severalstudiesduringthepastfewyearshaveunderscoreditscentralroleinmeta-bolicpathologies[5–7].ThisinvolvementofNF-kBinmetabolismandinflammationappearstoreflectthecloseintegrationandcoevolutionofnutrient-andpathogen-sensingsystems[6].Theabilitiestoendurestarvationandmountaneffectivedefenseagainstinfectionareamongthemostbasicrequirementsforspeciessurvival[6].Mountingaprotectiveimmuneresponse,however,isbioenergeticallycostly[5–7].Thus,fromametabolicper-spective,theneedtoprotecttheorganismfrompathogenscompeteswiththeneedtomaintainmetabolichomeostasis[6].Thispresentsafundamentalphysiologicalchallenge,whichhasbeenmetinevolutionthroughthecloseintegra-tionandcoordinationofimmuneandinflammatoryresponseswiththeregulationofmetabolism[6].
Thisconceptiseloquentlyillustratedbytheanatomicalcolocalizationofkeyconstituentsoftheimmuneandmet-abolicsystemsinlowerorganisms[6].InDrosophilamel-anogaster,immuneandmetabolicfunctionsarecontrolledbythesameorgan,thefatbody,whichisequivalenttothe
mammalianliverandadiposetissue,andalsothehemato-poieticandimmunesystems[6](Figure1).Theintimateassociationbetweenmetabolismandinflammationisalsoevidentinthearchitecturalorganizationofmetabolicorgans,suchastheliverandadiposetissue,inhigherorganisms.Here,macrophagesandotherimmunecellsareincloseproximitytometaboliccells,namelythehepa-tocytesandadipocytes[6](Figure1).Adipocytesandmacrophagesalsosharemanypathwaysandsignalingmodules[6],includingpattern-recognitionreceptorssuchasToll-likereceptors(TLRs),whichinduceimmunityandinflammationinresponsetopathogen-associatedproductsanddangersignals[8,9](Figure1).Notably,TLRfamilyreceptors,suchasTLR2andTLR4,whichrecognizelipo-proteinsandlipopolysaccharidesinbacterialwalls,alsosensenutritionalsignalssuchasfattyacids[5–7,10].Reciprocally,signalingpathwaysthataretraditionallyassociatedwithmetabolismcanaffectimmuneresponses(Figure1);metabolichormonessuchasleptinandadipo-nectinalsoregulateimmunefunctions[6,11]andlipid-sensingnuclearreceptorssuchasperoxisomeproliferatoractivatedreceptor(PPAR)andliverXreceptor(LXR)familymemberscansuppressinflammation[6].Theendo-plasmicreticulum(ER)isanothercrucialsitewheremet-abolicandinflammatorypathwaysconverge[5–7,12].Hence,nutrientscaninduceinflammatoryresponsesandpathogenscanevokeandregulatemetabolicsystems[6].Thisbiologicalconfigurationisparamountforthemainte-nanceofenergyhomeostasisandproperfunctioningoftheimmunesystem,butalsorepresentsamajorpredisposingfactorfortheonsetofmetabolicdiseases[5–7].
