IHC - Wholemount of Caenorhabditis elegans embryo labelling RNA polymerase II CTD repeat YSPTSPS with . Sample was incubated with primary antibody (1/100 in PBS + 3% BSA + 0.1% Triton-X 100) for 24 hours at 4&C. ab150113, an Alexa Flour& 488-conjugated goat anti-mouse IgG polyclonal (undiluted) was used as the secondary antibody.

stained HeLa cells. The cells were 100% methanol fixed (5 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (, 1&g/ml) overnight at +4&C. The secondary antibody (green) was ab150113 Alexa Fluor& 488 goat anti-mouse IgG (H+L) used at 1&g/ml for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43&M.The negative control (inset) is a secondary-only assay to demonstrate low non-specific binding of the secondary antibody.

The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (, 1&g/1x10^6 cells) for 30 min at 22&C. The secondary antibody used was Alexa Fluor& 488 goat anti-mouse IgG (H&L) at 1/2000 dilution for 30 min at 22&C. Isotype control antibody (black line) was mouse IgG1 [ICIGG1] (, 2&g/1x10^6 cells) used under the same conditions. Unlabelled sample (blue line) was also used as a control.&Acquisition of &5,000 events were collected using a 20mW Argon ion laser (488nm) and 525/30 bandpass filter. This antibody gave a positive signal in HeLa cells fixed with 4% paraformaldehyde (10 min)/permeabilized with 0.1% PBS-Tween for 20 min used under the same conditions.

Immunocytochemistry/ Immunofluorescence
iPS derived neurons (35 days) were fixed in 4% formaldehyde (10 min) and then incubated in 1% serum, 0.1% triton, 0.1% BSA in PBS for 30 mins to permeabilise the cells and block non-specific binding. The cells were then dual stained (simultaneous method) with antibody ab18207 (BIII Tubulin at 1/100 dilution) and Vglut1 (green) for 1h at room temp. Secondary antibodies used were Donkey anti-Rabbit IgG H&L Alexa 555 (ab150062) (red) and donkey anti mouse IgG 488 at 1:250 dilution and incubated for 1h at room temp. Total cells were stained with DAPI (blue). Positive BIII-Tubulin staining was observed in processes (green).
